Method Validation and Uncertainty Estimation for Total Phosphorus Determination in Animal Feed Using UV-Vis Spectrophotometer

Objective: The study presents the validation protocol for determination of total phosphorus content at parts per million (ppm) (μg/L) levels in animal feed by a UV-Vis spectrophotometer. Methods: measured absorbance solutions against blank solution was 400 nm with A combined ammonium heptamolybdate tetrahydrate and monovanadate blue used as coloring reagent detection. This method validated evaluation statistical parameters such linearity, sensitivity, limits detection (LOD) quantification (LOQ), precision, accuracy, measurement uncertainty using matrix (MB) standard. Results: Instrumental Detection Limit 0.066 ppm Quantification 0.22 ppm, respectively, while recovery repeatability percent were 101.15% 0.11%, respectively. However, linearity this 0.1 to 30 ppm. 2.82%, following Commission Regulation (EC). Conclusion: estimated protocol, found meet imposed performance criteria, procedure intended use.
